鼻粘膜におけるInsulin-Like Growth Factor-I (IGF-I) の
mRNAの発現

要 旨：IGF-Iが鼻粘膜内で産生されているのかを確認するため，IGF-IのmRNAの発現を，re-versetranscription-polymerase chain reactionをもちいて検討した。対症としては，鼻ポリープ，鼻アレルギー患者の下鼻甲介，及び肥厚性鼻炎の患者の下鼻甲介もちいた。さらに，鼻ポリープにおけるIGF-IのmRNAの発現が，局所適用性副腎皮質ステロイドであるプロピオン酸フルチカゾン（FDP）の投与によりその発現が抑制されうるかを半定量的RT-PCR法を用いて検討した。鼻ポリープおよび鼻アレルギー患者の下鼻甲介粘膜でのIGF-IのmRNAの発現を認めたが，非アレルギー肥厚性鼻炎患者の下鼻甲介粘膜では認めなかった。FDPによるIGF-IのmRNAの発現抑制実験にて，コントロールにIGF-Iの発現を認めるも，FDP下で培養した検体では，発現を認めなかった。

The Expression of Insulin-Like Growth Factor-I(IGF-I)
mRNA in Nasal Mucosa

Abstract: We consider insulin-like growth factor-I(IGF-I) is a growth factor of nasal polyps. To investigate expression of IGF-I mRNA in nasal polyps, reverse transcription-polymerase chain reaction (RT-PCR) was used for 10 cases of nasal polyps. 4 of 10 cases showed expression of IGF-I mRNA. And we used RT-PCR to investigate expression of IGF-I mRNA in the inferior turbinates of allergic patients and non-allergic patients. 3 of 4 cases of allergic and none of 3 cases of non-allergic patients showed expression of IGF-I mRNA. These results indicate that IGF-I has relation to the growth of nasal polyps and nasal allergy. This is the first report showed expression of IGF-I mRNA in nasal polyps and inferior turbinates of nasal allergy.
In recent years, topical corticosteroid therapy has been used for treatment of nasal polyposis. We examined by semi-quantitative RT-PCR whether topical corticosteroid FDP suppresses the expression of IGF-I mRNA in nasal polyps and showed that in one case FDP suppressed the expression of IGF-I mRNA.
