Immunoelectron Microscopic Localization of Sulfite 0xidase
in Eosinophil Leukocytes of Rat Bone Marrow and Intestinal Mucosa

Abstract: We studied the localization of sulfite oxidase [E.C. 1.8.3.1.] in eosinophils of rat bone marrow and intestinal mucosa Using an immunoelectron microscopical technique. Ultra-thin sections were immunostained for this enzyme by a protein A-gold technique. We observed immature eosinophil leukocytes at various developmental stages in bone marrow and the mature cells of the intestinal mucosa. Gold particles, showing the antigenic sites for the enzyme, were exclusively confined to the specific granules and mitochondria. In immunocytochemical control sections, no gold particles were observed. Quantitative analysis of the gold labeling showed that labeling density (gold particles/μu) of the specific granules was 3 times higher than that of the mitochondria. Analysis of the labeling of both specific granules and mitochondria during the maturation of the cells showed that the labeling density increases as the cells mature.
