Immunohistochemical Localization of Copper-Zinc and Manganese
Superoxide Dismutases in Human Tissues

Abstract: Superoxide anion radical which is produced in cells is considered to cause deleterious effects unless scavenged by superoxide dismutase (SOD). Immunolocalization of copper-zinc (FuZn) and manganese (Mn) S0D was studied by histochemical staining in formalin-fiixed human tissues. Staining by the indirect immunoenzyme method was performed using rabbit anti-human SOD antisera. The optimal dilution was 1:10,000 forboth anti-CuZnSOD and anti-MnSOD arti-sera. The high dilution of the antisera enabled a high resolution of immunostaining with a minimal background. The results were compared with those previously obtained from rat tissues. Human pancreatic exocrine cells, renal tubular epithelial cells and cardiac myocytes were intensely stained for MnSOD. The CuZnSOD reactivity was intense in human gastrointestinal and bronchial epithelium but not in cardiac myocytes or pancreatic islets nor in hepatocytes, as was in the case of rat tissues previously described. Although the staining profile of both SODs in human tissues was somewhat different from that in the rat tissues, there was a general trend of high density staining of either or both SOD types in epithelial linings and metabolically active sites.
